BUILDING YOUR OWN SIMPLE ONE LITRE FERMENTOR


The building of a simple one litre laboratory scale fermentor for teaching and even research gives the students a real hands on experience and understanding the application of principles of fermentor. The student will find this exercise challenging especially in ensuring aseptic integrity and anaerobic techniques.

It is recommended that the one litre culture vessel be obtained from Quickfit. The lid should be equipped with a number of port holes to accomodate:
1 Ph and temperature sensors or other probes
2 Sampling and nutrient feed tube
3 Aeration port
4 Thermistor probe

IMPORTANT NOTE

Since this fermentor will be based on magnetic stirring for mixing, make sure the glass base can withstand repeated sterilizations in autoclaves and do not warp! If warping and permanent deformation occur then proper mixing by magnetic bar will not be possible.

Heating of the fermentor can be obtained by using thermistor probe to a magnetic stirrer heater. A Whatman Data plate will be most suitable for this purpose and comes attached with its own thermoprobe.

Filtration of air is by using various PTFE bound membrane hydrophobic filter

It is recommended a simple rotameter be attached to regulate the air flow from aquarium diaphragm air pumps

Use tygon tubings for all purposes
